用热休克法诱导虹鳟四倍体最佳参数的研究Optimal parameters for tetraploidy induction in rainbow trout Oncorhynchus mykiss by heat shock

摘要:
采用热休克法抑制受精卵第一次卵裂诱导虹鳟Oncorhynchus mykiss四倍体,对影响热休克处理效果的3个参数:处理温度(26~31℃)、处理持续时间(3~16 min)、处理起始时刻(3 h 15 min~6 h 50min)进行了研究。结果表明:在授精后3 h 45 min,于31℃下热休克处理5、6、7 min,或30℃下处理7、10 min,或29℃下处理10 min,或28℃下处理16 min,都能成功地诱导出虹鳟四倍体胚胎,四倍体的诱导率为13%~33%,发眼期相对存活率为19.52%~96.54%。
